In the Phase 2 DAVIO 2 trial, a single dose of EYP-1901 maintained stable vision and retinal anatomy, reduced treatment burden, and controlled retinal fluid and fluid fluctuations for up to a year, with results comparable to aflibercept every 8 weeks in patients previously treated neovascular age-related macular degeneration (nAMD), according to a presentation at the Angiogenesis, Exudation, and Degeneration 2026 virtual meeting.
EYP-1901 is a bioerodible intravitreal insert that provides sustained release of the tyrosine kinase inhibitor vorolanib for at least 6 months. Vorolanib blocks intracellular VEGF receptor signaling and inhibits IL-6–mediated pro-inflammatory signaling via JAK1.
DAVIO 2 compared a single dose of EYP-1901 (2 mg or 3 mg) with aflibercept administered every 8 weeks in patients with previously treated nAMD. Patients with high baseline fluid (central subfield thickness [CST] >350 µm, IRF >25 µm, RPED >400 µm) were excluded.
The trial met its primary endpoint, with BCVA statistically noninferior after a single EYP-1901 dose versus aflibercept every 8 weeks, with stable vision and CST.
More than 60% of EYP-1901–treated eyes remained supplement-free through month 8. Baseline retinal fluid was low and comparable across arms, and 1 dose of EYP-1901 controlled retinal fluid through month 14, with results comparable to aflibercept every 8 weeks.
When fluid instability was measured using a fluctuation index, the 3-mg dose of EYP-1901 showed lower variability at months 8 and 14 than aflibercept.AI-assisted OCT analysis was used to identify anatomical biomarkers beyond CST.
Phase 3 trials of EYP-1901 are ongoing, with topline data from the LUGANO trial expected mid-2026.
